An O-ring issue has allowed sand to make its way into my pool. No a lot, but enough that over time I have “backwashsed” unknown amounts that was once part of my 100 lb. media filter. When I pop my multipurpose valve off to add sand how does one know when to say ‘when’?

SINCE you don’t know how much sand has been lost, there isn’t any way that I know of. My suggestion would be to remove the rest of the sand in the filter and put 100 lbs of new sand in the filter.
