I am purchasing this piece, but how is it attached into the filter assembly? Is it threaded and basically screws onto the base or would I need to glue the piece to the base?

This is a slightly different version of a DE filter. Normally the manifold is on the top of the grids. This one is on the bottom. Both versions are held together by either a double or ( in your case ) a single rod. There is no need for glue.
