we have an above ground 28' round pool with a pentair water sand filter system ; when the filter is running a water leak generates from the valve flange/plastic clamp area - is there a specific size O ring that we should replace?

Deb, You will need to replace the TA30D,35D,40D,50D,60D Valve Body O-ring (350) to keep the filter from leaking.

I'M looking for a sand filter and pump system for a 600-1200 gallons of salt water more or less to keep lobsters alive. Will this work?

This system would be more than enough to circulate and filter 600-1200 gallons of water.
